When is the best day to go to the show? The info website says that the first day (5th April) is a Happy View Day (for preview) but what does that mean?

How many times is it recommended to go to the show during the week? 1 day or 2 days?

And what does combined ticket with public transport mean? To and from where?

I am going on Wednesday. It's a sort of trade day, the entry is twice the normal rate for a short day, which keeps many away, but if you want to get good photos and move around freely it's a good day. I will be there till Friday afternoon, although I have done the show comfortably arriving at 11.00 on Friday, leaving 14.00 on Saturday.
If you are staying in the centre of Essen then the metro is an excellent way to get to the Messe, I think it will cover the train from Dusseldorf airport as well.

My best advice is not to walk around the show with Delwyn Mallett unless you want to hear him a) moan about his hip being painful, b) recount how he could have bought "that" pre-war Alfa Romeo for £1000 which is now worth £5 million but turned it down because it didn't run very well, or c) hear him waxing lyrical over yet another piece of rusty Italian shite.

Oh, and anyone can get in on Wednesday – see Nick's comment above about cost and benefits... You'll LOVE the show. I've been going for about the last 20 years and find it hard to miss.

How quiet is the Wednesday compared to the Thursday and Friday? We've accommodation in Duisburg from Wednesday to Saturday so we were going to do the Wednesday, Thursday, Friday and then head away Saturday morning to check out Classic Remise and maybe the Siku museum as they're kinda in the area and thought they'd be quieter because everyone would be at Essen instead...

Say you were going just for a single day, is there anything at Essen that shouldn't be missed ?

You could see everything in one day but it would be a rush and you wouldn't take it in or have time to stop and chat, two days is a minimum and three makes it a much more relaxing visit.
